Three experiments were carried out on different cultivars of cowpea (Vigna unguiculata L. Walp) during two summer seasons 2017 and 2018. The first study was conducted in the molecular biology laboratory, genetics department, Faculty of Agriculture, Assiut University., Assiut, Egypt., on seven cowpea cultivars to investigate the proportion of genetic relationship among seven cultivars of cowpea ”local and foreign cultivars” by revealing of molecular markers variation using RRAPD-PCR assay. The second and third experiments were carried out in the experimental farm of the vegetable department, Faculty of Agriculture, Assiut University, Assiut, Egypt. The aim of second investigation was to study the effect of three different irrigation intervals treatments (well-irrigated, moderate- stressed and drought-stressed) i.e., (10, 20- and 30-days intervals irrigations) and four cultivars of cowpea on growth, flowering, yield, protein content (%) and water use efficiency (WUE). The aim of third investigation was to estimates of green pods & dry seed yieldand quality in three cultivars of cowpea. Molecular markers and genetic variation revealed by RAPD- PCR in seven cowpea (Vigna unguiculata (L.) Walp.) cultivars. RAPD results showed that a total number of 56 DNA fragments were amplified, ranged in size between 1637 bp and 163 bp, while the total polymorphic bands number was 45 and the percentage of polymorphism was 80.3 %. Five cultivars (Cream-12, Chinese red Blackeye crowder, Brown crowder, and Cream-7) appeared to have whether positive or negative markers with different molecular weight, while Azmerly and Dokki-331 cultivars did not produce any specific bands (positive or negative markers). The dendrogram of RAPD data showed that the 7 cultivars of cowpea were grouped in 6 clusters based on genetic similarities ; the highest value of similarity was 86.6 % between Azmerly and Dokki-331 cvs., while the lowest was between Cream-12 and Blackeye crowder with a value of 54.2 %. The obtained results suggest that RAPD markers were better linked to the source of the cultivar and to the extent of seed crowding in pod trait, whiletheseedcolourandthegrowthhabittraitsmightormightnot show correlation with the molecular data. Impact of three gradient irrigation levels and four cultivars of cowpea on growth, yield and protein content. This trait gave the earliest to flowering with irrigation treatment T3 (30 days intervals), with values of (61.7 and 60.9 days to flowering) in both seasons respectively. Dokki-331 cv. was the earliest with values of (56.5 and 56.3 days to flowering) in both seasons respectively. Also, the interaction (Dokki-331 cv. x irrigation T3), gave the earliest to flowering with values of (54.03and 53.57 daysto flowering) in both season respectively.
