Résumé
Pigeon pea (Cajanus cajan (L.) Millsp.) is an important grain legume of the semi-arid tropics. It provides protein rich food. Pigeon pea being recalcitrant species, In vitro regeneration of pigeon pea through organogenesis was achieved from the field grown healthy auxiliary buds as explants of nine cultivars. Under the influence of variable concentrations of two cytokinins namely BAP and Kinetin. Shoot induction of variable morphology, was observed for all the cultivars and the hybrid GRPH-1 and GRPH-2 record highest regeneration (100 %). BAP was found to be most effective overall mean (72.83 %), Elongation of shoot was achieved on MS medium supplemented with BAP in combination with NAA and GA3. The combination of 2 mg/l BAP + 1 mg/l NAA + 2 mg/l GA3 was found to be most effective it increases 2 cm height compare to the other combinastion. The elongated shoots were successfully rooted on MS medium containing different auxins. Among them IBA 1 mg/l induce maximum frequency of rooting for all the cultivar over all mean (74.81 %) fallowed by IAA of overall mean (68.82) . On the other hand molecular confirmation studies using 12 SSR primer screened on the 9 genotypes and their tissue culture regenerated plant. 7 markers (CcM0008, CcM0021, CcM0057, CcM0121, CcMO133, CcMO262, CcM0268) showed monomorphism/homozygous bands it indicates that tissue culture-raised plants were genuinely true-to-type to the mother plant. further Two SCAR markers i.e. SCAR-704 and SCAR-N-18 that are linked to Fusarium wilt and SMD are amplified with a product size of 840 bp and 704 bp respectively. susceptibility reaction were screened on the tissue culture regenerated plants.
