Descriptif
A large number of genes undergo changes in their expression profile to impart drought tolerance in a tolerant genotype. A few of these would be key genes governing the expression of a larger set of genes and hence it is necessary to identify and understand regulation of genes that are differentially expressed during drought stress condition. Previously, we have generated 6053 ESTs from 8 different cDNA SSH libraries from the parents (drought tolerant ICC4958 and drought susceptible 1882) and extreme Recombinant Inbred Lines (RILs) exposed to terminal drought conditions during flowering stage by dry down method. The present study was tartgeted to explore potential candidate genes that can confer terminal drought tolerance by using comparative study of 1) 1CC4958 in control and drought stress (182 unigenes) 2) ICC 4958 and ICC 1882 under drought stress (830 unigenes) 3) Exteme RILs of high root and low root biomass lines under drought stress in comparison with control were studied (189 unigenes). In addition, validation of few candidate genes thorugh northern (23) or qRT-PCR analysis (10) and isolation of one full length gene (CarPSCS), expression study under various hormonal and abiotic stresses, cloning, transformation in Arabidopsis thaliana, isolation of upstream sequence (promoter) and in silica analysis for identifying stress responsive elements was performed
