Natural products can not only be used as highly effective and low-toxic insecticides and herbicides,but also an important source of innovative drug molecules to treat some serious human diseases.Therefore,the research on chemical biology of natural products will become a emerging filed with promising development prospect in the global pharmaceutical industry.In recent years,it has become more and more difficult to obtain natural products with novel structure and remarkable activity from microbial secondary metabolites in common habitats,so studies on fungi from special environmental sources have attracted extensive attention of scholars.In this thesis,two endophytic fungi from desert plants and one endophytic fungus from Harmonia axyridis were investigated with the aim of discovering compounds with new structures and efficient pharmacological activities.In the experiment,the secondary metabolites arised from the above-mentioned fungi were separated by using of various modern sparation methods.Finally,26 compounds were isolated and identified from the fermentation extracts of the three endophytic fungi,including 7 new compounds and 19 known compounds.16 compounds were obtain from the solid fermentation products of two desert plant endophytic fungi,Neocamarosporium sp.HM 175 and Alternaria sp.HM 213.1-1~1-5 were isolated from Neocamarosporium sp.HM 175,and 2-1~2-11 were isolated from Alternaria sp.HM 213.Among the 15 compounds,compounds 1-1,2-1,2-2 and 2-3 are new compounds.Compound 1-1 possesses a rare [6/6/6/6/5] tetracyclic cage anthrone analogs,and compounds2-1,2-2 and 2-3 are a pair of mixes with pyrrolidine diketone framework,respectively.The known compounds include two ketones(1-3 and 2-11),four coumarins(2-4,2-5,2-6 and 2-7),a mixed terpenoid(2-8),two perylene quinone(2-9 and 2-10)and three esters(1-4 and 1-5).10 compounds were obtained from the solid fermentation products of the insect endophytic fungus Fusarium equiseti,of which 3-1,3-2 and 3-3 were new compounds.Compound 3-1 is a pair of mixture with decalin and pyrrolidine diketone framework.Compound 3-2 is a [6/6/6/5] four-ring system with decalin skeleton,belonging to equisetin.3-3 is highly degraded sterol compound with characteristic side chains of common ergosterols.Equisetin compounds(3-4,3-5)and Fusarisetin compound(3-6)are characteristic metabolites of Fusarium equiseti.Compounds 3-4(equisetin)and 3-5(epi-equisetin)are a pair of differential isomers.Compound 3-6 has a [6/6/5/5/5] pentacyclic framework,3-7 and3-8 belong to the sterols compounds,3-9 and 3-10 belong to the esters compounds.14 compounds were tested for PTP1 B inhibitory activity in vitro.The results showed that compound 1-1、1-2、2-6、2-8、3-4、3-5 and 3-7 exhibited certain inhibitory activity against PTP1 B.The activity tests of these compounds provide potential drug lead compounds for the development of novel PTP1 B inhibitor
