The search for alternatives to solve the problems caused by environmental stresses is considered one of the major challenges of the national agribusiness. Currently, the effects caused by drought, low temperatures and high salinity in the soil have been investigated and several genes related to these characteristics have been isolated, characterized and used in plant breeding. The analysis of responses to stress in plants is an important route for discovering genes that confer stress tolerance and use them in breeding programs. Use of proteomics techniques, provide a broad overview of plant responses to stress at the protein level. Among the various plant species studied, the palm Opuntia cochenillifera Salm - Dyck., A forage of the Cactaceae family, presents relevant genetic characteristics related to high resistance under high temperature and low humidity, being able to survive for long periods of drought. Because of these characteristics we have decided to pursue a proteomics study of this plant under water stress conditions. To achieve this we compared protein extracts from leaves structures (cladodes) of the palm under normal conditions and under water stress, using uni-dimensional and bi-dimensional gel electrophoresis. A comparison of two-dimensional gels allowed the identification by MALDI TOF / TOF of protein common to both treatments and proteins that are differentially expressed between treatments. In this study, 20 protein spots were identified with differential expression and were classified in the following categories according to their possible physiological function in plants is divided into the following categories : 1) these related to photosynthesis (largest subunit of RUBISCO and Pyruvate Phosphate Dikinase - PPDK). 2 ) these related to energy storage (vicilin and legumin precursor), 3) glycolytic pathway (Fructose Bisphosphate Aldolase isoenzyme), 4) Antioxidants proteins (Nucleoside diphosphate kinase and NDPK Halocide Dehalogenase - Hydrolase), 5) these involved in signal transduction (such as Ligand Protein Nucleotide rich Leucine in Adenylate Kinase GTP dependent binding protein - MYA2, Pump Vacuolar proton ATPase, GTP binding protein activated by ethylene and aminocyclopropano carboxilato oxidase, 6) Signaling as α/β hydrolase, and 7) hypothetical or putative proteins with no described function. Our results strongly suggest that were first identified proteins involved in response to water stress in cactus.
