Résumé
Dwarf materials and drought-resistant cultivar Pubing 143 deprived from common wheat (Triticum aestivum)×Agropyron cristatum were used to analyze dwarf gene and identify drought resistance by morphology, chemical marker and SSR makers. The results are as following:Dwarf materials were developed by sexual cross among several parents, and their heredity backgrounds are complex. According to analysis of pidegree, it cannot be confirmed whether dwarf genes in common use are in parents. Among these parents, there are dwarf materials, and .Known molecular markers were used to eliminate dwarf genes in common use, it can be preliminarily confirmed that dwarf genes in common use were not in dwarf material 3648-3652 by contrast PCR products of dwarf material with control. Dwarf gene shows GA insensitivity through GA reaction identification. Alien chromosomal fragments were screened by SSR makers, 13 of 332 pairs primers can amplify PCR products same as A. cristatum. This result confirmed that the dwarf germplasm contained alien chromosomal fragments introgressed from A. cristatum.26 typically dwarf individual plants and 21 typically high individual plants from F2 population of 3648-52×Chinese Spring cross were selected to amplify by PCR. 10 of 26 dwarf individual plants could be amplified PCR products same as A. cristatum Z559, and 21 high individual plants have no such products. This result confirmed that 10 dwarf individual plants contained alien dwarf genes from A. cristatum. PCR products same as A. cristatum Z559 cannot be observed in the rest of 16 dwarf individual plants, dwarf trait maybe caused by other dwarf genes. Therefore dwarf material 3648-3652 contains more than one dwarf genes, a new dwarf gene is from A. cristatum among them. Plant height of F1 generation of 3648-52×Chinese Spring cross is 77.6cm, near to value of medium parents. Through dividual plant analysis, plant height of F2 population showed normal school. This result was in agreement with the result that material 3648-3652 contains more than one dwarf genes.Through identification of drought-resistance in seedling, plant survival rate is 68.75%, and Pubing 143 shows better drought resistance,is a drought-resistant cultivar.
