Broad bean (Vicia faba L), an important legume for human and animals, is rich in protein and is widely used throughout the world. These days new varieities resistant to diseases, cold stress, drought, and salt stress are developed using biotechnological methods. Very few studies are available on this subject in broad bean. The study aimed to develop a shoot regeneration method, more suitable for genetic transformation of broad bean. The seeds of Filiz 99 and Eresen 87 varieties were pretreated with water, liquid MS medium without sucrose, sucrosed water and liquid MS medium containing sucrose for 24 h. Thereafter, plumular tips and embriyonic axis were cultured on MS medium containing different concentrations of BAP-NAA. Maximum shoot regeneration was recorded on plumular apices on MS medium containing 0,25 mg/l BAP and 0,25 mg/l NAA. After 5 weeks the shoots were rooted in MS medium containing 1 mg/l IAA. In the second experiment, embryos of two cultivars were pre treated with 10 mg/l BAP for 6 days. Thereafter, plumular shoot tips obtained from the embryos were cultured on MS medium containing different concentrations of BAP and NAA. Maximum shoot regeneration was recorded on the explants pretreated with 10 mg/l BAP followed by culturing on MS medium containing 0.25 mg/l BAP and 0.25 mg/l NAA. The regenerated shoots were rooted on MS medium containing 1 mg/l IAA. The plants from both experiments were transferred to the greenhouse for flowering and seed set.
